Expression of hTR and hTERT in Human Breast Cancer

摘要 目的研究人端粒酶逆转录酶(hTERT)、人端粒酶RNA亚基(hTR)和预后指标包括肿瘤的大小、分级、结节状态和患者年龄间的联系。方法RT-PCR检测92例人乳腺恶性肿瘤和10例良性肿瘤的hTERT和hTR的表达。结果hTR和hTERT的表达分别为62例(67.4%)和76例(82.6%),与<40岁的人群比较,>40岁的人群hTR表达更显著,良性肿瘤中没有1例表达hTR,6例表达hTERT。hTR和hTERT的表达与肿瘤的分级、分期、淋巴结转移和病理分型呈显著相关,与肿瘤大小没有相关性,在76例hTERT阳性的患者中,均有不同程度的hTR表达阴性,在hTR阳性的患者中没有hTERT阴性。结论hTERT和hTR的表达同乳腺癌具有相关性;hTR比hTERT更能反映乳腺癌的发生情况。 Objective To study the relationship between telomerase sub-units of hTERT and hTR and the prognostic indicators including tumor's size and grade, nodal status and patients' age. Methods The expressions of h TR, hTERT, and β-actin (as a housekeeping) genes were determined by RT-PCR in 92 breast cancer samples and 10 benign tumor samples. Results The expressions of hTR and hTERT were found in 62 (67.4%) and 76 (82.6%) samples,respectively. There was a significant relation between hTR gene expression and the age at diagnosis in patients under 40, when compared with the patients older than 40 years. However, hTERT gene expression had no such relation. There was no expression of hTR gene,but the expression of hTERT gene in 6 samples of the benign tumors. The hTR and hTERT gene expression were closely associated with pathology, grade, stage and lymph node metastasis, but had no relationship with the size of breast carcinoma. The expression of hTR was negative with different degrees in 76 cases of hTERT positive expression,but there was no hTERT negative expression in 62 hTR positive cases. Conclusion The expression of hTR and hTERT has correlation with breast carcinoma. The hTR expression probably plays a greater role in mammary tumorogenesis than hTERT expression.
