氧化苦参碱对大鼠血管平滑肌细胞钙化抑制作用的研究 被引量:3

摘要 目的观察氧化苦参碱(Oxymatrine,OMT)对血管钙化的影响,探讨血管钙化的发生机制。方法以β-甘油磷酸盐诱导大鼠主动脉平滑肌细胞(VSMCs)钙化,OMT干预。采用磷酸苯二钠法测定碱性磷酸酶(ALP)活性,Von Kossa染色鉴定细胞钙化,比色法测定细胞钙含量,硫代巴比妥酸法测定细胞丙二醛(MDA)含量。结果β-甘油磷酸盐可诱导VSMCs发生钙化,钙化组细胞总钙含量、ALP活性和丙二醛含量明显增高,OMT呈浓度依赖性地降低细胞总钙含量、ALP活性和丙二醛含量,VSMCs的钙化程度亦明显减轻。结论OMT能有效抑制β-甘油磷酸盐诱导的大鼠主动脉VSMCs钙化,其机制可能与减轻细胞的氧化损伤有关。 Objective To explore the effect of oxymatrine(OMT) in vascular calcification in rats and its mechanism. Methods Rat calcific vascular smooth muscular cells (VSMCs) were induced by β- glycemphosphate, interfered with OMT. The total Ca^2 + contents in VSMCs were measured by colorimetric method; the alkaline phosphatase(ALP)activity in VSMCs were determined by phenyl diphosphate- 2- sodium; MDA content were measured by TBA method, calcification were observed in cultures by Von Kossa stain. Results β -glycemphosphate could induce calcification in VSMCs. The total Ca^2 + content, ALP activity and MDA content increased significantly in VSMCs.After the treatment of OMT, the calcification in VSMCs was attenuated and the total Ca^2 + content 、ALP activity and MDA content decreased in VSMCs in a concentration-dependent manner. Conclusion The results indicated that OMT can significantly attenuate the calcification in VSMCs. The possible mechanism is that it lessen the oxidative damage of the cells.
