摘要
目的观察中药复方TAP对胰岛素抵抗(IR)大鼠糖耐量、过氧化物歧化酶(SOD)、丙二醛(MDA)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)等指标的影响。方法SD雄性大鼠随机分为正常对照组和IR组,正常对照组予普通饲料,IR组予链脲佐菌素+高脂饲料,45d后测定糖耐量。选取IR组中糖耐量异常者随机分为模型组,二甲双胍(Met)组,TAP112、225、450mg/kg3个剂量组。给药60d后,检测糖耐量及血清SOD、MDA、IL-6、TNF-α含量。结果与模型组相比,TAP112mg/kg组在0、60min2个时间点和TAP225、450mg/kg组在0、30、60、120min4个时间点血糖明显降低,TAP450mg/kg组SOD值明显升高,TAP225、450mg/kg组MDA和IL-6值明显降低,TAP112、450mg/kg组TNF-α值明显降低,差异均有统计学意义(P<0.05或P<0.01)。结论TAP可显著改善IR状态,其机制可能与增强抗氧化能力、减轻炎症反应有关。
Objective To observe effects of TAP on glucose tolerance and serum level of SOD, MDA, IL-6 and TNF-α in IR rats. Methods Male SD rats were randomly divided into normal group with common feed and insulin resistance group with small-dose STZ plus high-fat feed. Glucose tolerance were measured after 45 days fed. Unnormal glucose tolerance rats were randomly divided into the model group, mefformin treatment group and TAP of three dosages(112,225,450 mg/kg) treatment groups. After 60 days treatment, glucose tolerance, serum level of SOD, MDA, IL-6 and TNF-α were measured. Results Compared with the model group, the level of blood sugar at 0, 60 min in TAP112 were decreased significantly mg/kg group and at 0, 30, 60, 120 min in TAP225,450mg/kg groups (P〈0.05, P〈0.01), SOD level in TAP450 mg/kg group were increased significantly (P〈0.05), MDA and IL-6 level in TAP225,450 mgkg group was decreased significantly (P〈0.05, P〈0.01), TNF-α level in TAP112,450 mg/kg group was decreased significantly(P〈0.05, P〈 0.01). Conclusion TAP can improve insulin resistance, and its mechanism may be concerned with enhancing antioxidative ability and inhibiting inflammation reaction.
