p73、p63及p53基因表达与喉鳞状细胞癌预后相关性分析 被引量:3

摘要 目的探讨p73、p63和p53蛋白在喉鳞状细胞癌发生、发展中的作用与意义,为喉鳞癌的临床诊断及治疗提供新的思路,并为基因治疗的探索提供理论依据。方法采用免疫组织化学(S-P法)检测p73、p63、p53蛋白在50例喉鳞状细胞癌、10例癌旁组织及10例正常喉黏膜中的表达,分析3种基因蛋白与喉鳞癌的临床病理特征之间的关系。结果(1)p73表达与局部淋巴结转移成正相关(P<0.01)。p73阳性表达的患者生存率相对较低(P<0.05)。(2)p63表达与临床分期成负相关(P<0.05),与其他临床特征及生存率无关(P>0.05)。(3)p53表达与各临床特征均无关,p53阳性表达的患者生存率相对较低(P<0.05)。结论(1)喉鳞状细胞癌中的p73过表达可能与局部淋巴结转移有关。p73阳性表达可能是患者预后不良的指标。(2)p63过表达的肿瘤恶性度可能相对较低,p63的表达与患者预后无直接联系。(3)p53阳性表达的患者预后较差。(4)提示p53家族可能和喉鳞状细胞癌形成的早期阶段有关。 Objective To investigate the possible effects of p73,p63 and p53 during the progression of laryngeal squamous cell carcinoma,and to provide value clues for searching and developing new approaches to clinical diagnosis and therapy for larynx squamous cell carcinoma. Methods Immunohistochemistry method was used for detecting p73,p63 and p53 protein expression in the tissues of laryngeal squamous cell carcinoma (50 cases), atypical hyperplasia of larynx mucosa(10 cases) and normal mucos related to larynx squamous cell carcinoma(10 cases). Results (1)Expression of p73 was significantly and positively associated with the lymph node metastasis (P〈 0.01). The patients with p73 positive expression always had lower survival rate than those with p73 negative expression.(2)Expression of p63 was significantly and negatively associated with clinical stage(P〈0.05),and had no relations with prognosis of patients.(3) Expression of p53 had no distinct difference with clinical parameters.The patients with p53 positive expression always had lower survival rate than those with p53 negative expression (P〈0.05).Conclusions (1)It is possible that the overexpression of p73 in laryngeal squamous cell carcinoma is correlated with lymph node metastasis.P73 expression,possibly be a marker of poor prognosis.(2)It is possible that tumors with overexpression of p63 are correlated with relatively lower malignancy.No distinct relations are found between expression of p63 and prognosis.(3)P53 expression is an important marker of poor prognosis.(4)It is possible that p53 family is correlated with formation of larynx squamous cell carcinoma in the early stage.
