摘要
为了探讨知母宁对急性缺氧性肺动脉高压的影响及其作用机理。对知母宁对急性常压低氧肺动脉高压SD大鼠模型平均肺动脉压(mPAP)、血浆丙二醛(MDA)水平及肺组织磷脂酶A_2(PLA_2)活力的影响进行实验观察,并以加内皮素-1(ET-1)和血小板活化因子(PAF)分别进一步对其作用机理进行探讨。结果表明缺氧大鼠mPAP升高的同时,伴有肺组织PLA_2活力和血浆MDA水平的升高。知母宁可拮抗缺氧、ET-1及PAF所致的mPAP增高,但对血浆MDA水平和肺组织PLA_2活力增加无影响。由此证实,知母宁确有降低缺氧性肺动脉高压的作用,其机理可能与拮抗ET-1和PAF的作用有关。
To determine the influence of chinonin on acute hypoxic pulmonary arterial pressure in SD rats and investigate its action mechanism. 35 SD rats were divided into five groups at random; 6 of them in control group; 6 in hypoxia group; 13 in the group of endothelin - l(ET- 1) added chinonin; 5 in the group of platelet activating factor ( PAF) added chinonin . Their mean pulmonary arterial pressure ( mPAP ) was measured and the MDA in their plasma and the activity of PLAo of their lung tissue were detected.
Results showed that mPAP^MDA in plasma and activity of PLA2 of their lungs were increase when they inhaled 7% of oxygen. Chinonin could prevent the increase of mPAP caused by hypoxia and could antagonize the action of ET - 1 and PAF which could promote mPAP. But chinonin had no influence on the increase of MDA in the plasma and PLA2 of the lungs when hypoxia.
All these indicated that Chinonin could reduce the hypoxic pulmonary arterial pressure, couldn' t influence the normal mPAP. It may be acted by blocking the role of ET - 1 and/or PAF or others. The definite mechanism need be studied further.
