糖尿病大鼠下颌骨成骨细胞体外特性的研究 被引量:14

Effect of alloxan-induced diabetes mellitus on the differentiation of osteoblasts obtained from rat mandible in vitro

摘要 目的:研究体外培养糖尿病大鼠下颌骨成骨细胞生长分化的特性。方法:20只6周龄雌性SD大鼠随机平均分为2组,实验组腹腔注射四氧嘧啶(200mg/kg),对照组注射生理盐水。模型建立成功10d后,取下颌骨行成骨细胞培养。MTT法、PNPP法、考马斯亮蓝法、放射免疫法及钙结节茜素红染色检测细胞的增殖能力、蛋白校正ALP含量、骨钙素水平及矿化能力。结果:实验组2d、4d、6dMTT相对比值高于对照组(P<0.05),3d、7d、14d、21dALP含量低于对照组(P<0.01),28d总骨钙素水平以及钙结节的数量及面积均低于对照组(P<0.01)。结论:急性糖尿病早期促进体外培养大鼠下颌骨成骨细胞的增殖,抑制其基质成熟和矿化,可建成具有内在缺陷的成骨细胞模型。 Objective: To analyse the biological behaviour and differentiation of osteoblasts obtained from diabetic rats' mandibles. Methods: 20 female Sprague-Dawley rats were assigned to two groups equally and randomly: diabetic group induced by alloxan(200mg/kg) peritoneal injection and control group injected physiologic saline. The rats were killed and mandibles were harvested 10 days after the successful induction of diabetes for osteoblasts culture. Comparison of cell biological activity was made between the two groups using MTT, PNPP, BGP content radioimmunoassay and alizarin bordeaux stain of calcified nodules. Results: MTT value (2d、4d、6d) of diabetic group was significantly higher than the control group (P〈0.05), yet ALP(3d、7d、14d、21d) and BGP(28d) value were on the contrary (P〈0.01). Conclusion: Early acute diabetes mellitus may promote osteoblasts' proliferation but inhibit extracellular matrix maturation and mineralization in vitro optimal microenvironment. Diabetic osteoblasts with intrinsic defects model may be developed.
