摘要
为研究流式细胞术定量分析肿瘤细胞增殖细胞核抗原的表达,采用双参数流式细胞术检测方法和单克隆抗体免疫荧光染色技术定量分析增殖细胞核抗原表达。结果Ec8712、8113、7901三株细胞的PCNA平均表达量分别为31.56%,78.14%和74.35%,双参数检测结果表明,PCNA主要在S期和G2M期高表达。双参数流式劝胞术检测方法结合单克隆抗体免疫荧光染色技术能较准确地定量分析肿瘤基因蛋白的表达量。
In order to quantitatively analyze the expression of proliferating cell nuclear antigen(PCNA), a dual parameters flow cytometry (FCM) and immunofluorescence staining technique were used. The PCNA average expression of human esophageal cancer cell line Ec8712, oral squamous cell carcinoma cell line 8113, and gastric cancer cell line 7901 cells were 31 56%, 78 14%, and 74 35%, respectively. The result of dual parameters detecting measurement showed that the high expression of PCNA was in S phase. The PCNA expression was accurately analyzed by this method. It might be a new useful means for cancer diagnosis and prevention.
