日本血吸虫成虫RNA的快速分离纯化

RNA ISOLATION FROM ADULT SCHISTOSOMA JAPONICUM BY SINGLE-STEP METHOD

应用异硫氨酸胍-酚-氯访一步法提取日本血吸虫总RNA，在1h左右即可提取纯度高、完整性较好的RNA。该方法简单易行，既经济又省时，对来源不易的少量组织尤为有效。通过对血吸虫成虫RNA变性琼脂糖凝胶电泳，观察到日本血吸虫大亚基rRNA存在体内缺口现象，随核糖体小亚基RNA一道电泳迁移。

Total RNA was isolated from adult S.japonicum by single-stepnol-chloroform micture.The mehtod with TRIzol reagent provides both high yield and purity of undegraded RNA preparation and allows isolation of RNA in one hourlSince the single-step method is simple and time saving,it is particularly useful for processing large numbers of samples and for isolation of RNA from small quantities of samples.Analysis on agarose gel electrophoresis indicated that mouse live RNA revealed two major bands i.e.28s and 18s,while S.japonicum adult RNA one 18s band only.
