热休克对不同细胞RNA和HSP70mRNA合成功能的影响

摘要 45℃加热10分钟可以杀死50%的中国白鼠卵巢(CHO)细胞和57%的Hela细胞。热休克也强烈地抑制RNA的合成,使细胞内RNA水平急剧降低。hathernBlot对HSP70mRNA的分析表明热休克并不影响细胞内已有的少量HSP70mRNA。但热休克后HSP70mRNA的合成会迅速增加。这将改变细胞的生长调控和促进受损细胞的快速增长。 The test of clonecounting living cells showed that 50% of Chinese Hamster Ovary (CHO) cells and 57% of Hela cells were killed when the cells were heated in 45℃ watr bath for 10 minutes. Heat-shock strongly destroyed RNA in cells and inhibited the transcriptive function of gene so that RNA content in cells decreased fast. The analysis of HSP 70 mRNA by Northern Blot showed that heat-shock did not degrade HSP 70 mRNA in two cells. The HSP 70 mRNA synthesis quickly increased after heat-shock. It shows that the inhibited HSP gene in normal growing condition is uninhibited. The transcription and trans-lation of the uninhibited HSP gene will undoubtedly change the growing regulation of cells and promote the proliferation of the injured cells fast.
