摘要
目的建立一种内毒素诱导的急性肺损伤(ALI)幼猪模型。方法健康雄性幼猪16头随机分为正常对照组6头、内毒素(LPS)组10头。正常对照组给予0.9%氯化钠溶液(10ml/kg);内毒素组缓慢泵注LPS(15μg/kg),机械通气条件下观察肺氧合等肺功能损害的进程。结果注射LPS后6h动脉血氧分压/吸入氧浓度(PaO2/FiO2)、肺泡-动脉氧分压差(A-aDO2)出现显著改变,12h达到ALI标准,LSP组注射LPS3h、6h、12h肺动态顺应性和气道阻力与注射前0h比较,差异有统计学意义(P<0.05),与对照组同一时间比较差异亦有统计学意义(P<0.05),支气管肺泡灌洗液分析及肺形态学出现显著异常。结论本法可诱导稳定的脓毒性ALI猪模型,供ALI病理生理机制及治疗方法研究之用。
Objective To establish a kind of models of acute lung injury (ALI) induced by endotoxin. Methods Sixteen healthy male piglets were randomly divided into 2 groups: normal group (n = 6 pigs) and LSP group (n = 10 pigs). The normal group were given injections of normal saline ( 10 ml/kg) and the LSP group a slow infusion of lipopolysaccharide (LPS) ( 15 ug/kg). Lung function damage process, such as lung oxygenation, was observed under the condition of mechanical ventilation. Results PaO2/FiO2 and A - aDO2 changed significantly at 6 h after LPS infusion, and reached ALI standards at 12 h. Significant anomaly was observed in bronchoalveolar lavage liquid analysis and lung pathomorphology. Conclusion By this method we can induce a stable septic ALI piglet model for the research purposes of ALI pathophysiological mechanisms and treatment.
