摘要
目的通过研究肠缺血再灌注损伤后血必净注射液对肠道免疫屏障中T淋巴细胞亚群中P53基因表达的影响,探讨在肠缺血再灌注损伤后血必净注射液对肠粘膜免疫屏障的作用机制。方法取方法:将Wistar大鼠60只随机分为2组,模型对照组(CON)和血必净注射液组(XBJ),再按实验时间分为24h、48h、72h组各10只。采用免疫组化双染色法分别观察造模后24h、48h、72h大鼠小肠粘膜固有层内CD4+、CD3+、CD8+T淋巴细胞数中P53基因表达的情况,比较实验组与对照组P53基因表达的差异。结果实验组与对照组相比较,第48h、72hCD4+、CD3+T淋巴细胞中P53基因表达对照组高于实验组(P<0.05,P<0.01);CD8+T淋巴细胞中P53基因的表达第24h、48h、72h两组均无差异性(P>0.05)。结论血必净注射液能够通过对肠壁免疫细胞凋亡基因P53表达的调控,实现对肠道免疫屏障的调节作用。
Objective To research the influence of Xuebijing injection on express of P53 of T lymphocytes and IgA plasmocytes of intestinal wall after intestinal ischemia-reperfusion injury,so as to explore the mechanism of Xuebijing injection on immune barrier of intestinal wall.Methods 60 Wistar rats were divided into two groups randomly.including the control model group and XBJ group.each group was divided into two subgroups（24h,72h respectively）.Each subgroup has 10 Wistar rats.The express of P53 gene of CD4^＋,CD3^＋,CD8^＋ T lymphocytes of ileal mucosal and laminapropria of the distal end of deal mucosal tissue were observed by immuohistochemical method,further more,the difference Between the control model group and XBJ group.were compared.Results The express of P53 of CD4^＋,CD3^＋ T lymphocytes in control group is higher compared with it in experiment group on the 48 and 72 hours（P〈0.05,P〈0.01）;The express of P53 of CD8^＋T lymphocytes in control group has no difference compared with it in experiment group（P〉0.05）.Conclusion Xuebijing injection can inhabit the express of P53 in gut immune barrier rises during the gut barrier injury.it may effect immune barrier of intestinal wall by this means.
