早实核桃外植体材料培养初步研究 被引量:2

摘要 以3个新疆早实核桃品种为试材,对外植体的消毒方法、取材时间、培养基和外植体类型等因子进行了初步研究。结果表明:两次升汞消毒法能增强茎尖的消毒效果;以嫩枝茎段为外植体时,适宜的取材时期为6月,以腋芽饱满半木质化的培养效果较好;适宜的培养基类型是DKW和1/2MS,嫩枝茎尖的培养效果要好于茎段;3个品种的初代培养效果存在差异。 With three Xinjiang precocious walnut varieties as experimental materials, the influence of sterilization ways of explants, time of material - taking; and types of medium and explants were preliminary studied. The results showed that two steps of sterilization with mercuric chloride enhanced the sterilization effect for shoot tip ; the most suitable time for taking tender shoots with full axilla buds and semi - lignification as explants was in June; the shoot tip had better culture results than tender shoots; and that DKW and 1/2MS medium were both fitting for the precocious walnut explants in vitro culture ; differences of the primary culture effect of three varieties was observed.
