补肾B方对老龄SD大鼠细胞因子表达的调节作用

Regulation Effect on the Expression of Cytokins in Aged Rat of SD With Recipe B of Restoring Kidney of Traditional Chinese Medicine

目的:研究中医补肾药方(B方)对老龄SD大鼠细胞因子表达的影响。方法:把12月龄雄性SD大鼠随机分为实验组和对照组,实验组给予灌服补肾B方药液,90天后,处死动物,摘取脾脏,提取RNA。同时采用3月龄雄性SD大鼠作为年轻对照组。采用美国Affymetrix公司RayBio大鼠细胞因子抗体芯片试剂盒,检测细胞因子表达水平,并进行对照比较。结果:15月龄大鼠比3月龄大鼠的IL-10表达增加,而CNTF、β-NGF、TNF-α、IL-1α、IL-1β、IL-6的表达下调;补肾B方灌服SD大鼠后,下调了IL-10的表达,同时上调了CNTF、CINC-3、IL-1α、IL-1β、IL-4、LIX(Leptin、MIP-3α、β-NGF和TNF-α细胞因子的表达水平。结论:机体细胞因子的表达随着增龄而变化,中医补肾活血化痰可调整衰老机体以IL-10升高为特征的免疫衰老状态。

Objective ：To investigate effect on the expression of cytokins in aged rat of SD with recipe B of restoring kidney of traditional Chinese medicine（TCM）. Methods ：8 of male rats of Sprague dawley（SD） of 3mo. with clean and healthy were selected as young control group; 16 of male rats with 12mo. with clean and healthy were randomly allocated to experimental group I , 8 ,and 8 as older control group. Recipe B for restoring kidney composed of gouqi, shouwu, yinyanghuo,dancan and juhua etc. of TCM,was made to the liquor with lg/mL,respectively. Experimental group I were given Recipe A liquor in the light of 5g/kg for 90 days. The RNA was extracted from spleen taken out of rats, respectively. The cytokines were determined with Rat Cytokines Antibody Array I by Affymetrix of USA. Results ： Cytokines in rats of 15mo. are changing with aging, namely elevation of IL - 10 and decline of CNTF, β - NGF, TNF - α, IL - 1 α, IL - 1 β and IL - 6 by comparison with young control grou. The down of regulation of IL - 10 and he up regulation of CNTF, CINC - 3, IL - 1α, IL - 1β, IL - 4, LIX, Leptin, MIP - 3α, β - NGF and TNF - α by comparison with older control group, after treatment with Recipe B liquor. Conclusion ：Cytokines in aged rats are changing with aging;Integration restoring kidney with activation of blood circulation and dispelling phlegm of TCM may regulate immunosenescence in aged characteristic with elevation of IL - 10.
