人参皂苷Rb1对体外条件下小鼠骨髓间充质干细胞和粒-巨噬系祖细胞增殖的影响 被引量:5

摘要 目的:探讨人参皂苷Rb1在体外培养条件下对小鼠骨髓间充质干细胞(MSC)生长的影响。方法:设不同终浓度人参皂苷Rb1的细胞培养体系与对照,检测MSC集落生成率,M竹比色法测定MSC集落培养体系细胞的活力。结果:在培养体系中人参皂苷Rb1浓度为10μg/mL时,MSC集落产率和MTT吸光值与对照组无明显差异(P〉0.05);25~100p.g/mL的范围内,MSC集落培养体系细胞集落产率和MTT吸光值显著高于对照组(P〈0.05),其中浓度为50μ/mL时效应最明显(P〈0.01);在200μg/mL时,相应检测指标减低,但并未显示抑制作用(P〉0.05)。结论:人参皂苷Rb1在一定浓度范围内对体外培养条件下的小鼠骨髓MSC具有生长促进作用,但在高浓度时,其促进作用反而减弱。 Objective: The present study was designed to investigate the effects of ginsenoside of ginsenoside Rb1 on the growth of routine bone marrow mesenchymal stem cells(MSC) in vitro. Methods: The colony yields of MSC were assessed in the murine bone marrow cell cultures at various concentrations of ginsenoside Rb1 and in the control groups. The MTT assay were used to determine the cell viability in MSC cultures. Results: There was not any significant difference in the colony yields of MSC between the 10 μg/mL ginsenoside Rb1 groups and controls. The colony - forming efficiencies and cell viability of MSC culture were significantly increased in the MSC cultures with 25μg/mL to 100μg/mL ginsenoside Rb~. The maximal effect was observed at 50μg/mL. However, the decreased percentages of corresponding measurements of MSC at 200μg/mL ginsenoside Rb1 were lower than control groups. Conclusion: ginsenoside Rb1 over a range of concentrations has enhancing effects on the in vitro growth of murine bone marrow mesenchymal stem cells.
