中药早期干预对幼年狼疮鼠肾组织TGF-β NF-κB表达的影响

Influence of Early Treatment of Chinese Herbal Medicine on the Expression of TGF-β NF-κB in Kidney of Juvenile Lupus Mouse

目的:探讨解毒祛瘀滋阴中药早期干预对幼年MRL/lpr鼠肾损害的保护作用。方法:2月龄雄性MRL/lpr小鼠50只及2月龄雄性C57BL/6小鼠10只,随机分成低、中、高3个不同剂量组、阳性对照组、治疗前组及正常对照组6组研究。治疗前组直接取标本;其余5组分别给药30天后,取标本。检测肾脏TGF-β1mRNA、NF-κB p65表达;肾脏病理组织形态的改变。结果:治疗前组、阳性对照组肾组织TGF-β1mRNA、NF-κB p65表达较正常对照组明显增强;经中药早期干预的高剂量组则表达明显减弱,差异有显著性(P<0.001)。经中药早期干预的高剂量组肾小球系膜细胞、基质增生较阳性对照组明显好转;中剂量组较阳性对照组减轻;低剂量组无明显改善。结论:解毒祛瘀滋阴中药早期干预,能明显降低肾组织TGF-β1mRNA的表达;能抑制肾组织中NF-κBP65的表达;并且能明显改善MRL/lpr小鼠狼疮性肾炎的病理损害。

Objective： To discuss the protection of Chinese herbal medicine that removing toxic substances and blood stasis and nourishing Yin on the Juvenile MRL/lpr mouse. Methods ： Applied 50 pieces of 2 - month - old male MRL/lpr mouse and 10 pieces of 2 - month - old male C57BL/6 mouse as the observation objective, which was randomly divided into six teams including three treating teams of high, middle and low dosages, positive contrast team, pre - treatment team and normal contrast team. Do not give the pre -treatment team any medicine and immediately take sample ＇after di- viding team; for other five teams, the sample will be taken after being given medicine for 30 days. Detect the expression of TGF - β1mRNA and NF -κB p65 in nephridial tissue. Under optical microscope, observe the variation of pathology in kidney. Results： The kidney organization of TGF- β1mRNA and NF- κB 1365 in positive contrast team, pre- treatment team has strong expression compared with the normal contrast team. The expression of that of those in treating team with high dosage was significant weakened, while compared with the positive contrast team （ P 〈0.001 ）. And, the hyperplasia of cell and stroma of mesangium in the treating team of high dosage after the early treatment of Chinese herbal medicine was obviously well - treated, compared with the positive contrast team. And the treating team of middle dosage was eased, but the treating team of low dosage has no obvious improvement. Conclusion： The early treatment of the Chinese herbal medicine can obviously weaken the expression of TGF - β1mRNA and NF - κBP65 in kidney tissue. It can also obviously ease the pathology damage of the MRL/lpr juvenile mouse lupus- prone nephritis.
