湖北地方小麦品种大头黄低分子量麦谷蛋白亚基基因的分子克隆

摘要 采用PCR方法从湖北地方小麦品种大头黄(ZM11217)中克隆得到1个低分子量麦谷蛋白亚基(LMW-GS)基因LMW-ZM11217。该基因具有LMW-GS基因的典型结构特征,编码区长度为870bp,编码288个氨基酸。推导的氨基酸序列比较结果表明,LMW-ZM11217与Glu-A3和Glu-D3位点控制的基因具有较高的相似性(最高相似性分别为82%和83%),而与Glu-B3位点控制的基因具有较低的相似性(最高相似性为74%)。 Using PCR method,the coding sequences of a low molecular weight glutenin (LMW-GS) gene, LMW-ZM11217, was isolated from Hubei wheat landrace accession ZM11217. It had a coding region of 870 bp, and encoded a mature protein of 288 amino acids, LMW-ZM11217 was a typical LMW-GS gene. Comparison of the deduced amino acid sequence showed that LMW-ZM11217 had higher similarity with genes at Glu-A3 and Glu-D3 locus (with the highest similarity 82% 83%, respectively), whereas it had lower similarity with genes at Glu-D3 locus (with the highest similarity 74%).
