灰树花斜面母种培养基的优化 被引量:1

Optimization of Mother Strain Slope Media for Grifola frondosa

摘要 试验在传统培养基基础上,分别从葡萄糖单因素培养、合成培养基正交培养与半合成培养基正交培养3个角度或层次对灰树花(Grifola frondosa)母种斜面培养基进行了优化探讨。结果表明,适合灰树花菌丝生长的最佳斜面培养基配方为土豆200g、麸皮100 g、玉米面100 g、板栗壳100 g、葡萄糖20 g、林地土20 g、蛋白胨1 g、琼脂20 g、CaCl2 0.1 g、KH2PO41 g、FeSO4 1 g、MgSO4 0.5 g、ZnSO4 0.01 g、水1L、pH值6.0。该配方既可为灰树花母种的制备提供依据,又能为灰树花的大面积栽培以及为实现工业化大生产而进行的液态发酵培养奠定基础。 Optimization of mother strain slope media for Grifola frondosa was researched by single factor culture with glucose, synthetic media culture and semi-synthetic media culture based on the traditional mother strain slope medium. The results showed that the best mother strain slope medium components (MSM) were 200g of potato, bran 100g, corn powder 100g, the chestnut hull 100g, glucose 20g, wooden soil 20 g, peptone 1g, agar 20 g,CaC12 0.1g, KH2PO4 1g, FeSO4 1g, MgSO4 0.5g, ZnSO4 0.01g, water 1L, pH6.0, which was suitable for the culture of Grifola frondosa, It was helpful for the preparation of mother strain slope medium and the foundation of solid culture and liquid submerged fermentation of Grifola frondosa.
