摘要
对桑黄菌原生质体进行He-Ne激光诱变处理,经过3代筛选,筛选出了5株变异菌株JS1至JS5。5株变异菌株多糖发酵产量比出发菌株均有所提高,多糖含量也都发生了变化。酯酶同工酶分析表明变异菌株酯酶同功酶图谱发生了变化;传代培养表明变异菌株遗传性较稳定;正变异率估算表明He-Ne激光诱变桑黄菌原生质体正变异率较高,适宜用于桑黄菌原生质体诱变。
Protoplast from Phellinus igniarius was irradiated by He- Ne laser. Five mutant strains which were JS1, JS2, JS3, JS4, JS5 were selected after three generations. The polysaecharide content was also increased. Results showed that esterase spectrogram of five mutant strains was changed by analysis of esterase isoenzyme electropheresis spectrogram, the mutant strains have good hereditary stability from generation to generation. After valuation of mutation rate, it proved that the positive mutation rate of mutation by He - Ne laser on Protoplast from Phellinus igniarius was higher, This irradiation method was suitable for Protoplast from Phellinus igniarius.
