荧光染色法观察LED活化MPPa介导的光动力作用诱导卵巢癌细胞凋亡 被引量:3

摘要 目的:探讨LED活化MPPa介导的光动力作用诱导人卵巢癌细胞凋亡的光化学生物学效应。方法:应用Hoechst33258细胞核荧光染色法观察LED活化MPPa介导的光动力作用对人卵巢癌细胞凋亡的影响。结果:LED活化MPPa介导的光动力作用组出现核凝聚、凋亡小体的人卵巢癌细胞株COC1/DDP细胞明显多于单纯LED辐照组、单纯MPPa光敏剂处理组和假辐照组,而单纯LED辐照组、单纯MPPa光敏剂处理组和假辐照组三对照组中出现核凝聚、凋亡小体的细胞非常少见。结论:LED可以作为一种新型的光源活化MPPa等光敏剂介导光动力作用从而有效诱导人卵巢癌细胞株COC1/DDP细胞凋亡。 Objective: To observe the photochemical biological effect of Lighting Emitting Diode (LED )- activated MPPa on the apoptosis of ovarian carcinoma celia. Methods: A special lighting emitting diode equipment was employed to investigate the photechemical biological effect of MPPa 4h after photodynamic therapy in a hmnan ovarian carcinoma cell line COC1/DDP by Hoechst33258 fluorescent staining analysis. Results: Hoechst33258 staining showed that the morphological changes in the nuclear chromatin of LED - activated MPPa in COC1/DDP were extraordinarily significant. In the control groups, the nuclei were stained a less bright blue and the color was homogeneous. After being irradiated by LED with MPPa for 4h, the blue emission light in apoptotic cells was much bright than the control cells. Condensation of chromatin and nuclear fragmentations could also be found in many treated cells and some of them formed the structure of apoptotic bodies, which is one of the classic characteristics of apoptotic cells. Conclusion:Lighting Emitting Diode (LED) - activated MPPa could effectively induce apoptosis of human ovarian carcinoma cells and Lighting Emitting Diode (LED) will become an alternative light source for photodynamic therapy.
