摘要
目的:考察不同显色方法对紫外-可见分光光度法测定知母总皂苷含量的影响。方法:采用紫外-可见分光光度法测定知母总皂苷含量,分别用两种显色方法,显色剂方法1为香草醛-冰醋酸法。即香草醛-冰乙酸:高氯酸:冰乙酸(2:8:50)。检测波长为545nm。显色方法2为浓硫酸法,即用浓硫酸:甲醇(1:4)。检测波长为270nm。结果:采用方法1显色,知母皂苷A3在0.058~0.58mg范围内线性关系良好(r=0.9999),知母皂苷样品加样回收率分别为100.19%。RSD为2.81%;方法2显色,知母皂苷A3在0.0128~0.64mg范围内线性关系良好(r=0.9996),知母皂苷样品加样回.收率分别为98.50%,RSD为5.04%。结论:综合精密度实验、重复性实验、准确度实验等指标。方法1明显优于方法2.且该方法简便、准确、灵敏、重现性好。
Objective: To investigate the effects of two different chromogenic agents on determination of the content of total sap- onins in Anemarrhena by UV spectrophotometry. Methods:Total Anemarrhenasaponin were determinated by two different chromo- genie agents, namely method one, vaniUin - glacial acetic acid perehlorle acid glacial acetic acid (2: 8:50)at 545 nm;method two, sulfuric acid- methanol( 1:4 ) detected at 270 nm. Results:Using method one, the linear range of Anemarrhenasaponin A3 was 0. 058 ~0. 58 mg(r =0. 9999). The recovery rate was 100. 19% ,RSD2. 81% ; using method two,the linear range of Anemarrhe- nasaponin A3 was 0. 0128 - 0.64 mg( r = 0. 9996 ). The recovery rate was 98.50% , RSD4. 96%. Conclusion : From these index, the first method was better than the second method. This method was simple ,accurate and the reproducibility was good.
