异氟醚对大鼠急性心肌缺血再灌注损伤的保护作用 被引量:1

摘要 目的观察异氟醚预处理对大鼠急性心肌缺血再灌注损伤中心肌热休克蛋白27(HSP27)和核转录因子-κB(NF-κB)表达的影响,探讨异氟醚抗心肌缺血再灌注损伤的可能作用机制。方法健康成年Wistar大鼠30只,随机分为3组,假手术组(Ⅰ组)吸入纯氧1h,24h后仅行左冠状动脉套线而不阻断血流150min;缺血再灌注组(Ⅱ组)吸入纯氧1h,24h后结扎左冠状动脉前降支阻断血流30min,再灌注120min;异氟醚实验组(Ⅲ组)吸入2%异氟醚和纯氧1h,24h后处理同I/R组,建立在体心肌缺血再灌注动物模型。用免疫组化对心肌HSP27和NF-κB进行检测。结果Ⅱ组、Ⅲ组心肌HSP27和NF-κB活性增高,再灌注120min后表达明显增加,预处理延迟相可上调HSP27表达,下调NF-κB表达,均有统计学意义(P<0.01)。结论异氟醚可通过抗氧化作用,增加HSP27的表达和减低NF-κB的活性发挥抗心肌缺血再灌注损伤,对心肌有明显的保护作用。
