桃核承气汤对蓄血证大鼠血管MMP-2,TIMP-2基因表达的影响

Effects of Taohechengqitang on Gene Expression of MMP-2 and TIMP-2 in Artery Vessels of Experimental Xuxuezheng Rats

[目的]观察桃核承气汤对蓄血证大鼠血管基质金属蛋白酶-2(Matrix Metalloproteinase-2,MMP-2)及金属蛋白酶-2组织抑制剂(Tissue Inhibitor of Metalloproteinase-2,TI MP-2)基因表达的影响,探讨此方的作用机制。[方法]半定量RT-PCR检测各组大鼠颈总动脉匀浆中MMP-2、TI MP-2基因的表达,进行分析。[结果]与模型组相比,药物组减少了MMP-2基因的表达,增加了TI MP-2的表达(P<0.01)。[结论]桃核承气汤能改变蓄血证大鼠动脉壁MMP-2、TI MP-2基因的表达,这可能是其主要作用机制之一。

[Objective]To investigate the effects of Taohechengqitang（THCQT） on the gene expression of Matrix Metalloproteinase- 2（MMP-2） and Tissue Inhibitor of Metalloproteinase-2（TIMP-2）in artery vessel of Xuxuezheng rats. [Methods] Half quantitative reverse transcription polymerase chain reaction（RT-PCR） was used to detect MMP-2 and TIMP-2 gene expression in artery vessel. They were analyzed. [Results]While compared with model group, MMP-2 gene expression levels in the THCQT group were lower, and TIMP-2 was higher （P〈0.01）. [Conclusion]THCQT can change the gene expression of MMP-2 and TIMP-2 in artery vessels of Xuxuezheng rats, which may account for the effects.
