明胶法测定精子顶体酶活性的研究

摘要 目的:探讨明胶底物膜法测定精子顶体酶活性相关技术的改进和质量控制。方法:①采用传统方法100℃溶解明胶,56℃铺片和改良方法50C溶解.35C铺片分别得100张明胶片.对比明胶片的质量。②比较不同密度精液在35张明胶片上涂片的试验结果.寻找合适的涂片密度。③比较新鲜胶片和储存2周的胶片在顶体酶释放阳性率和平均晕环直径两方面实验结果上的差异。结果:①改良方法和传统方法制得的胶片在合格率方面P〈0.05,差异有统计学意义。②密度(7~13)×10^6ml范围的精液涂片后试验结果疏密均匀。③新制和储存2周的明胶片在顶体酶释放阳性率、顶体酶释放平均晕环A径的比较中.均P〉0.05.差异无统计学意义。结论:经过改进明胶片的制作方法和对精液密度方面进行质控后.明胶底物膜法在实验结果上更可靠;明胶片能够储存使实验操作简便化,更能适合临床的应用。 Objective:To report a modified and quality controlled gelatin lysis test for measuring acrosin activity in semen. Methods:① The quality of 200 pieces of gelatin plates made half in traditional method (lyse gelatine at 100℃, spread out gelatine at 56C ) and half in modified method (lyse gelatine at 50℃, spread out gelatine at 50℃ ) was compared. ②Different density of 35 samples of semen were compared and elected for searching suitable density of semen in gelatin lysis test,③The differences of mean halo diameter and percentage of halo formation in 16 cases sample on gelatin plates made immediately and gelatin plates made two weeks ago were compared. Result: ① The difference was statistically significant between traditional method and modified method in making gelatin . (P〈0. 005). ② The density of semen limited (7- 13) × 10^6/ml is better. ③It was not statistically significant on mean halo diameter and percentage of halo formation between new gelatin plates and gelatin plates made two weeks ago. (P〉0.05). Conclusions:The result is reliable in using modified method in making gelatin plates and suitable density of semen.
