摘要
目的:分离、培养和纯化大鼠切牙根尖蕾细胞,观察Notch 1在根尖蕾细胞中的表达。方法:分离出生后7dSD大鼠下颌切牙胚,切取根尖唇侧部分,酶消化法原代培养,差别胰酶消化法纯化上皮细胞。角蛋白14和波形丝蛋白染色鉴定细胞来源。Notch 1免疫细胞化学染色。结果:原代培养的细胞经2~3次差别胰酶消化后可获得纯化的根尖蕾上皮细胞。免疫细胞化学染色角蛋白14阳性,波形丝蛋白阴性,并且在根尖蕾细胞中存在Notch1阳性细胞。结论:体外培养的根尖蕾细胞中存在Notch 1表达阳性的细胞,Notch信号途径可能参与了根尖蕾中干细胞增殖和分化的调控。
AIM: To isolate, culture and purify the rats'apical bud cells and observe the expression pattern of Notch 1 in the apical bud cells cultured in vitro. METHODS: We separated the lower incisor germs from postnatal 7 d SD rats, cut the labial apical part of these germs and cultured the mixed cells by enzymatic digestion. Then, the apical bud cells were purified by differential trypsinization and identitied by immuno -staining of cytokeratin 14 and vimentin. The Notch 1 expression was examined by immunocytochemistry. RESULTS: The apical bud cells were purified after 2 -3 times of differential trypsinization, and were positive stained for cytokeratin 14 and negative for vimentin. CONCLUSION: Notch 1 is positively expressed by some special apical bud cells cultured in vitro, which indicates that Notch pathway may be involved in the maintenance of the apical bud cells growth.
