兔外伤性增生性玻璃体视网膜病变中碱性成纤维细胞生长因子和增殖细胞核抗原表达的变化 被引量:1

bFGF and PCNA expression in rabbit traumatic proliferative vitreoretinopathy

摘要 目的探讨碱性成纤维细胞生长因子(basic fibrob-last growth factor,bFGF)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)在兔外伤性增生性玻璃体视网膜病变(proliferative vitreoretinopathy,PVR)中表达的动态变化及其相关性。方法采用原位杂交法和免疫组化法分别检测兔外伤性PVR模型bFGF和PCNA表达,染色结果行图像分析。结果伤后bFGF和PCNA表达呈现先升高后降低的动态变化,14d组阳性表达最高(平均积分光密度分别为18502.31±10822.69和89343.75±19686.71),与7d、21d、28d组比较差异有显著性;bFGF和PCNA表达之间有显著相关性(r=0.580,P≤0.01)。结论bFGF参与外伤性PVR增殖细胞的生长调控,与细胞增殖的动态变化呈高度相关性。 Objective Proliferative vitreoretinopathy (PVR) is a reactive process of the ocular tissue after perforating trauma, retinal detachment, and surgical manipulations. It is characterized by a proliferation of retinal pigment epithelium cells, neuroglial cells, and inflammatory cells on the retina, under the retina and in the vitreous cavity. Proliferative vitreoretinopathy (PVR) is still the most common cause of failure due to surgery for rhegmatogenous retinal detachment, despite the substantial effort that has been devoted to a better understanding and management of this condition during the past 25 years. Ohr study explores the dynamic expression and correlation between bFGF (basic fibroblast growth factor) and PCNA (proliferating cell nuclear antigen) in rabbit traumatie proliferative vitreoretinopathy (PVR). Methods Expression of bFGF mRNA in retina was detected by in-situ hybridization examination, and expression of PCNA in the retina was detected by immunohistoehemical analysis. The staining results were analyzed with an image anal- ysis system. Results After establishing an animal model for the expression of bFGF mRNA and the PCNA protein, there was an initial up-regulation and then a down-regulatlon, with the peak value in 14 d group (mean integrated optical densities were 18502.31±10822.69 and 89343.75±19686,71 separately), that was significantly different from those in the 7 d, 21 d and 28 d groups. There was a significant correlation between bFGF and PCNA expression (r=0.580). Conclusion bFGF takes part in the regulation of proliferative cells in traumatic proliferative vitreo- retinopathy (PVR), and is highly correlated with the dynamic changes of cell proliferation.
