摘要
目的探讨沙利度胺(Thd)对健康人外周血T淋巴细胞的免疫调节作用。方法健康人外周血T淋巴细胞经不同浓度Thd作用后,用MTT比色法检测其增殖情况,用流式细胞术检测其早期凋亡及CD28、CD152表达,用RTPCR检测其白细胞介素6(IL-6)、IL-10、肿瘤坏死因子α(TNF-α)mRNA表达水平。结果在体外,与阴性对照组相比,500μg/mlThd组可促进T淋巴细胞的增殖、早期凋亡,抑制T淋巴细胞CD28的表达。100μg/mlThd组可促进T淋巴细胞CD152表达。各剂量组均能抑制IL-6、TNF-αmRNA表达,100μg/ml和500μg/mlThd可增强IL-10mRNA的表达。结论Thd可以通过影响T淋巴细胞的增殖、早期凋亡以及CD152、CD28,IL-6、TNF-α、IL-10mRNA的表达,对健康人T淋巴细胞具有免疫调节作用。
Objective To study the immunoregulatory effect of thalidomide on human peripheral blood T lymphocytes. Methods T cells were treated by thalidomide with different concentrations. T cell proliferation was detected by MTT, and apoptosis, lymphocyte activation marker were analyzed by flow cytometry. The mRNA expression of interleukin (IL)-6, IL-10 and tumor necrosis factor (TNF)-α were measured by RT-PCR. Results Thalidomide 500 μg/ml not only promoted T cells proliferation but also caused their early apoptosis and inhibited the expression of CD3^+ CD28^+. Thalidomide 100 μg/ml promoted CD8^+ CD152^+ expression of T-cells as well. Thalidomide in the concentrations from 2.5 to 500 μg/ml inhibited the mRNA expression of IL-6 and TNF-α, but both of thalidomide 100 μg/ml and 500 μg/ml promoted IL-10 mRNA expression. Conclusion Thalidomide has the immunoregulatory effects on human T cells by promoting cell activation,proliferation, early apoptosis and regulating the expressions of some cytokines.
