连环恒温扩增技术研究进展
摘要
连环恒温扩增技术是针对靶基因的6个区域设计4种特异引物,利用一种链置换DNA聚合酶在恒温65℃左右保温几十分钟而完成的核酸扩增反应,通过检测反应过程中获得的副产品焦磷酸镁所形成白色沉淀的混浊度来判定扩增结果。由于其具有快速、简便、特异性好、成本低等优点,已经广泛应用于临床诊断、食品卫生检疫及基因芯片的开发;在细菌和病毒的检测方面对临床医学有重要意义。本文就连环恒温扩增的技术原理及其应用作一综述。
