摘要
目的观察替尼泊苷(VM-26)、依托泊苷(VP-16)及氟尿嘧啶(5-FU)对U-251人胶质瘤细胞及U-251胶质瘤干细胞的生长抑制作用,探讨胶质瘤干细胞对传统化疗药物敏感性.方法运用无血清培养法从人胶质瘤细胞株U-251中培养分离出胶质瘤干细胞,然后用MTT比色法测定氟尿嘧啶、依托泊苷、替尼泊苷对U-251胶质瘤细胞及U-251胶质瘤干细胞的增殖抑制作用,比较U-251胶质瘤细胞及U-251胶质瘤干细胞对这3种化疗药物的敏感性.结果U-251胶质瘤细胞对血浆峰浓度值的VM-26、VP-16和5-FU的敏感性明显高于U-251胶质瘤干细胞.结论对U-251胶质瘤的生长起关键作用的U-251胶质瘤干细胞,对血浆峰浓度值下的VM-26、VP-16和5-FU不敏感,胶质瘤的治疗应靶向胶质瘤干细胞.
Objective To study the sensitivity of U -251 glioma cells and glioma stem cells to some chemotherapeutic drugs (Vm -26, VP -16, 5 -FU) , so as to provide some effective clues for clinic chemotherapy. Methods Glioma stem cells were isolated from U - 251 human glioma cell line, MTT assay was performed to evaluate the inhibition effects upon the proliferation of U -251 glioma cells and glioma stem cells, and the sensibility of these drugs were compared. Result The inhibition rate of VM - 26, VP - 16, 5 - FU on glioma cells were evidently higher than glioma stem cells. Conclusion Glioma stem cells are more resistant to chemotherapy than glioma cells, the latter is the main target for chemotherapy.
