摘要
目的:探讨中药治疗对哮喘大鼠IL-13 mRNA和TNF-α mRNA表达及IgE含量的影响.方法:Wistar大鼠,随机分为中药治疗组(n=8),空白对照组(n=8),哮喘模型组(n=8),利用卵白蛋白(OVA)及氢氧化铝制作大鼠哮喘模型,在雾化激发期,中药组用杏仁、甘草等中药制成的溶液雾化大鼠,其余组用生理盐水代替.支气管肺泡灌洗液中细胞分类计数,采用免疫放射法、逆转录聚合酶链反应(RT-PCR)半定量法分别检测各组大鼠血清和肺泡灌洗液(BALF)中免疫球蛋白E(IgE)含量以及肺组织IL-13 mRNA和TNF-α mR-NA表达.结果:与空白对照组比较,哮喘模型组大鼠IgE含量、EOS计数、IL-13 mRNA和TNF-α mRNA的表达显著升高.与哮喘模型组比较,中药治疗血清IgE的含量、IL-13 mRNA和TNF-α mRNA的表达明显降低(P<0.05).结论:中药治疗可抑制IL-13和TNF-α mRNA合成,改善哮喘炎症状态.
AIM: To assess the modulation of IL-13 mRNA, TNF-α mRNA and IgE with Chinese herbal drugs in rats with bronchial asthma. METHODS:Twenty-four Wistar rats were randomly divided into 3 groups: control group, asthma model group, therapeutic group. Rat model of asthma was made by ovalbumin (OVA) and AI (OH)3. In the period of challenge, a Chinese herbal solution comprised with bitter apricot seed, liquoric root etc was used in therapeutic group. While in other groups normal saline was inhalated. Differential count of cells in bronchoalveolar lavage fluid was performed. Quantification of IgE in serum and bronchoalveolar lavage fluids (BALF) and IL-13 mRNA and TNF-α mRNA levels in lung tissues were conducted by immunoradiometric assay ( IRMA ) and RT-PCR. RESULTS : Compared to the controls, the asthma model rats had significant increases of serum IgE level, eosinophils percentage, and expressions of IL-13 mRNA and TNF-α mRNA. Compared to the asthma model group, the rats in the therapeutic group showed significant reductions of serum IgE and expressions of IL-13 mRNA and TNF-α mRNA. CONCLUSION: The Chinese herbal solution we used in this research could inhibit the expressions of IL-13 mRNA and TNF-α mRNA and improve the inflammation of asthma.
