细胞周期蛋白G_1参与孕激素对子宫内膜基质细胞增殖作用的研究 被引量:1

摘要 目的在体外培养条件下,研究孕激素(P)对子宫内膜基质细胞(ESC)的增殖作用和细胞周期分布的影响及细胞周期蛋白G1(cyclin G1)的表达情况。方法分离小鼠ESC进行原代培养,采用四甲基氨唑蓝法(MTT)检测P处理后ESC的增殖情况,用流式细胞仪检测细胞周期各时相的分布,用细胞免疫化学技术检测cyclin G1蛋白的表达情况。结果孕激素对ESC起增殖作用,在一定范围内呈时间依赖性。孕激素作用12 h后,S期细胞百分数明显上调。雌激素(E)、孕激素(P)、雌激素+孕激素(EP)联合分别刺激ESC 24 h,P组和EP组的ESC增殖,S期的细胞百分数升高,且cyclin G1蛋白表达阳性。E组与空白对照组(C组)的ESC增殖抑制,S期的细胞百分数维持低水平,且cyclin G1蛋白表达阴性。结论孕激素对ESC起增殖作用,引起细胞周期重新分布可能是孕激素促ESC增殖的机制之一。cyclin G1呈孕激素依赖性表达,可能参与孕激素对ESC的促增殖作用。 Objective It is to observe the influence of progesterone (P) on the proliferation, cell cycle distribution and Cydin Gi expression of endometrial stromal cells (ESC) in vitro. Methods Methyl thiazolyl tetrazolium method (MTT) was used to detect the cell growth status of ESC stimulated by P. Cell cycle distributions of ESC were analyzed by flow cytometry (FCM). Immunocytochemistry was used to measure the expression of cyclin G1 protein, Results Progesterone promoted the proliferation of ESC in a time-dependent manner as measured by MTT method. Progesterone increased the percentage of cells in the S phase after 12 h. There was no expression of cyclin Gi protein in group C and group E whose percentage of cells in the S phase was low. On the contrary, cyclin G1 highly expressed in group P and group EP whose percentage of cells in the S phase increased. Progesterone promoted cyclin G1 expression in cultured ESC when the proliferation of cells was significantly increased and cell cycle distributions were changed at the same time. Conclusion Progesterone has a potent proliferative effect on ESC, the mechanisms of which may be related to the induction of re-distribution of cell cycle. Cyclin G1 whose expression depends on progesterone may be helpful for the action of progesterone on the proliferation of ESC.
