肿瘤谱阵列芯片技术分析人体不同癌组织hHBrk1基因的差异表达 被引量:1

摘要 目的检测不同肿瘤及其癌旁正常组织中hHBrk1基因的差异表达,为进一步研究hHBrk1基因对肿瘤生物学过程的影响提供线索。方法以154对正常及肿瘤组织(19种不同人体组织)和10种肿瘤细胞株的cDNA的肿瘤谱阵列芯片(Cancer profiling arrayⅡ)为研究材料,采用Northern杂交方法分析目的基因表达水平的差异。根据阳性结果收集临床肿瘤组织样本,用RT-PCR检测hHBrk1基因表达,以验证芯片结果的可靠性。结果154对组织及10个肿瘤细胞株均表达hHBrk1基因。其中肺肿瘤组织高表达hHBrk1基因,与正常配对肺组织表达丰度差异有显著性(P<0.01);在肾癌组织中的hHBrk1基因表达丰度低于配对肾组织(P<0.01)。RT-PCR证实8临床肺肿瘤组织中5例高表达hHBrk1基因。结论肿瘤谱阵列芯片技术能够快速检测hHBrk1基因在不同组织中的表达差异,hHBrk1基因可能与肺癌及肾癌的发生、发展有关。 Objective To detect the expression profiling of the F-actin related gene hHBrkl in human tumors and matched normal tissues. Methods Cancer profiling array, contains of 154 matched eDNA pairs from 19 kinds of tumors and normal tissues and 10 kinds of cell lines, were hybridized separately with radio-labeled probes for the hHBrkl and housekeeping gene ubiquitin. RT-PCR assay was applied for conform the expression profiling of hHBrk1 gene in lung cancer tumors and matched normal tissues. Results hHBrkl gene was detectable in all of the tissues and cell lines. Significantly higher expression of hHBrkl was detected in lung cancer tissues, and inversely lower expressed level of hHBrkl was found in kidney cancer tissues compared with the normal tissues. Conclusion The Cancer Profiling array offers a new approach for detecting the expression of target gene. hHBrk1 gene might involve in the tumorigenesis and progress of lung and kidney cancer.
