摘要
目的探讨体外人巨噬细胞基质金属弹力酶(HME)对人胃癌细胞血管内皮生长因子(VEGF)表达的影响。方法体外培养人胃癌细胞SGC7901,并通过不同剂量HME蛋白干预胃癌细胞VEGF表达,HME干预剂量为1~5μg/ml,分别于干预后0、12、24、36h采用ELISA方法检测细胞上清中VEGF浓度变化并统计分析,同时设定对照组。结果干预36h后,HME干预剂量在1~5μg/ml时VEGF浓度均低于对照组,差异有显著性(P<0.05);对照组12h后VEGF表达明显高于0h,差异有显著性(P<0.05),在1、2、4、5μg/ml剂量组中,干预后12h与0h比较,差异无显著性(P>0.05)。结论HME蛋白浓度在1~5μg/ml范围内时在作用36h后能显著地抑制胃癌细胞VEGF表达。
Objective To study the effects of human macrophage metalloelastase on the expression of VEGF in gastric cancer cells. Methods To cultivate gastric cancer cells and interfere in the expression of VEGF in gastric cancer cells with different HME doses from 1 μg/ml to 5μg/ml, setting control groups meanwhile. Results The VEGF expression level after intervening for 36 h was reduced significantly compared with control group, P 〈 0.05, after intervening for 12 h ,the level of VEGF of control group increased significantly compared to 0 h ,P 〈0. 05. HME at 1,2,4,5 μg/ml had no significant differences between 0 h and 12 h,P 〉 0. 05. Conclusion HME at 1 - 5 μg/ml inhibited the expression level of VEGF significantly after intervening for 36 h.
