摘要
本文在模拟精练工艺条件下,用紫外吸收光度法和SDS-聚丙烯酰胺凝胶电泳技术,从精练时丝胶溶失速率、溶失方式和水解特性等方面作了初步研究。指出茧丝丝胶层中不同分子量的蛋白质组分在沿半径方向上的分布是不均匀的。精练时,茧丝外层易溶丝胶和内层难溶丝胶的溶失方式不同;在不同精练条件下,外层易溶丝胶的溶失方式也有差异;阴离子表面活性剂十二烷基硫酸钠(SDS)具有在脱胶过程中先促进丝胶溶失,后期又抑制丝胶溶失的双重作用。分子量较高的丝胶蛋白质组分水解稳定性较差,且肽链中间肽键易于水解断裂。
This paper, using ultraviolet absorption spectrophotometric determinater and sodiumdodecyl sulfate polyacrylamine gel electrophoresis,deals with the dissolution rate, the re-moval proceeding and the hydrolytic characteristic of sericin under the simulating scour-ing condition. It is found out that the sericin polypeptide are not well-distributed overthe sericin layer of silk filament. There is difference between the soluble sericin and theinsoluble one in the removal proceeding,which of the soluble sericin is not the same underdifferent conditions. Sodium dodecyl sulfate (an anionic surface active agent) can speedup the dissolution rate of sericin in the initial stage of scouring, and then, slow down thedissolution rate of sericin. The large sericin polypeptides are hydrolyzed readily, and theattack tends to take place in the middle of the peptides chains.
