摘要
目的:了解层粘连蛋白γ_2链基因(Lnγ_2)是否在正常大鼠肝组织中表达及其在肝纤维化过程中的作用。方法:以皮下注射四氯化碳(CCl_4)诱发大鼠肝纤维化模型,采用地高辛原位杂交技术对 Lnγ_2 mRNA 在肝组织中表达进行检测,并与Ⅳ型前胶原α_1链(α_1(Ⅳ))基因表达进行比较。结果:正常大鼠肝组织无 Lnγ_2 mRNA 表达,纤维化早期肝组织即有 Lnγ_2mRNA 的异常表达,其表达细胞肝内分布与α_1(Ⅳ)相似。结论:纤维化肝组织的间质细胞能表达 Lnγ_2基因,提示层粘连蛋白异构体基因表达在肝纤维化早期基底膜异常沉积中起一定作用。
Objective:To clarify the expression of the γ_2 chain of laminin(Lnγ_2)gene in normal and fibrotic rat liver.Method:Hepatic fibrosis model in rat was induced by administration of carbon tetra- chloride.The γ_2 chain of laminin gene expression in liver tissue was studied by in situ hybridization using digoxigenin-labeled DNA probe.Result:In normal liver,no positive cell can be found.In fibrotic liver; markedly enhancement of Lnγ_2 gene expression was determined mainly in mesenchymal cells.Conclusion: The γ_2 chain Laminin gene expression is activated in liver fibrogenesis.This result indicate that the ex- pression of isoform of iaminin in fibrotic liver may play an important role in the formation of basement member during the early process of liver fibrosis.
