宫宁颗粒对放置宫内节育器大鼠模型子宫血管构筑的影响 被引量:8

摘要 目的建立放置宫内节育器的大鼠模型,观察放置宫内节育器后大鼠子宫血管构筑的变化及宫宁颗粒的干预作用。方法Wistar雌性已生育大鼠56只,SPF级,造模后随机分为7组:空白对照组、假手术组、模型组、宫宁大剂量组、宫宁中剂量组、宫宁小剂量组和吲哚美辛(消炎痛)组,每组8只;造模后第31天,宫宁大、中、小剂量组分别给予宫宁颗粒配制液临床用量2、1、0.5倍灌胃,消炎痛组给予消炎痛配制液临床用量1倍灌胃,空白对照组、假手术组及模型组均灌胃同体积饮用水,连续给药7d后停药23d,再给药7d,于停药12h后,单宁酸-Fe法媒染子宫血管并观察血管构筑。结果模型组大鼠子宫各级血管扩张增粗,走行僵直,螺旋状和蜿蜒状走行消失或明显减少,血管形成的各级血管网密度增加,分布不均;消炎痛组、宫宁小剂量组子宫血管仍存在较明显的血管腔扩大、走行僵直和血管网密度增加,宫宁大、中剂量组子宫血管构筑的异常改变消失或明显改善。结论放置宫内节育器可影响大鼠的子宫血管构筑,宫宁颗粒较之消炎痛能明显改善子宫血管构筑的异常改变,这可能是其治疗宫内节育器引起的子宫异常出血机制之一。 Objective To investigate the effects of Gongning granula on uterine vessels formation in rats model with intrauterine device ( IUD ). Methods Fifty-six female post-labor rats of SPF grade were randomly divided into 7 groups, the normal control group, simulative operation group, model group, large, middle and small dosage Gongning group,indomethacin group,8 rats in each group. Rats in the latter 5 groups were made into IUD model by inserting homemade IUD into rats' uterus, and the same operation without laying IUD was given to rats in simulative operation group. Medication was started 31 days later, the large, middle and small dosage Gongning group was respectively treated with Gongning granula configuration solution 2, 1,0. 5 multiple clinical therapy dosage by gastric gavage, the indomethacin group with indomethacin configuration solution 1 multiple clinical therapy dosage, and to the other three groups equal volume of drinking water was given, After 7 days continuous gastric gavage 23 days later,the same medication was given. Twelve hours latter, uterine vessels formation was revealed by tannin-Fe lavement solution, Results The uterine vessel formation of rats in the model group showed that varices, tetanic shape, the density of all levels vessel net increased obviously and its twist,serpentine shape decreased markedly,while that in the large, middle dosage Gongning groups displayed absolute contrarily and fit like a glove with the normal control group, The uterine vessel formation of rats in the small dosage Gonguing group and indomethacin group also showed the same transformation as it in the model group. Conclusions To lay IUD into rats uterus could transform uterine vessel formation and Gongning granule could prevent that transformation completely than indomethacin,which is the mechanism of Gongning granule curing abnormal uterus bleeding induced by IUD.
