摘要
对蚶科三种贝类ITS2进行PCR扩增、测序及分析,用Mega3.1软件进行了系统发育初步分析。结果显示:毛蚶、泥蚶和魁蚶的ITS2大小分别为284~288bp、380bp、413~417bp,三种蚶的445个序列比对位点中,共有172个保守位点,245个变异位点,166个简约信息位点,78个单突变位点。系统发育分析结果显示,毛蚶和泥蚶先聚为一支,而后与魁蚶聚在一起。ITS2分析结果显示毛蚶和泥蚶种间的遗传关系较近。
Three species of Arcidae' s ITS2 are amplified, sequenced and analysed. With the software of Mega3.1, we analysed the development of phylogenetic. The results show that the size of Scapharca subcrenata' s ITS2 sequence is 284 -288bp; Tegillarca granosa' s ITS2 sequence is 380bp; Scapharca broughtonii' s ITS2 sequence is 413 -417bp. There are 445 sequences loci which contain 172 conserved loci;245 variable loci; 166 parsim-info loci and 78 singleton loci in the three species of Arcidae. The analytical result of phylogenetic development show that Tegillarca granosa and Scapharca subcrenatas are classified in one branch of Phylogenetic tree, then are classified with Scapharca broughtonii is in another branch, the analytical result of ITS2 show that the genetic relation between Tegillarca granosa and Scapharca subcrenatas are closed.
