培养基成分对桑树试管苗黄酮类化合物含量的影响

摘要 基于探讨适合桑黄酮类化合物合成的优化培养条件,以桑品种陕桑305为材料,研究了培养基成分及其添加量对桑树试管苗黄酮类化合物含量的影响。结果表明:当培养基中果糖的质量浓度为30g/L,氮元素浓度为60.02mmol/L,氨态氮与硝态氮物质的量比为0.3165,MnSO4·4H2O和ZnSO4·7H2O的质量浓度分别为11.15、4.3mg/L,激素质量浓度为0.1mg/LNAA(α-萘乙酸)、0.5mg/L6-BA(6-苄氨基嘌呤)、0.5mg/LKT(激动素)时,有利于桑树试管苗叶片中黄酮类化合物的合成;Mn2+和Zn2+浓度能影响苯丙氨酸解氨酶活性,当培养基中的MnSO4·4H2O和ZnSO4·7H2O分别为11.15、4.3mg/L时,酶活性最高。 Using mulberry variety Shansang 305 as experiment material, we investigated the effect of culture medium compositions on the flavonoids content in leaves of mulberry tissue culture seedling in this paper. Results showed that it was propitious for the mulberry seedlings to synthesize flavonoids, when the culture medium contained 30 g/L fructose, 60. 02 mmoVL nitrogen, 0. 316 5 nitrogen ratio of ammonia form to nitrate form, 11.15 mg/L MnSO4 · 4H2O, 4. 3 mg/L ZnSO4·7H2O, 0.1 mg/L NAA (α-Naphthylacetic acid), 0. 5 mg/L 6-BA (6-Benzylaminopurine), and 0. 5 mg/L KT (Kinetin). The result of experiment also indicated that Mn^2 + and Zn^2 + concentration can influence the activity of phenylalanine ammonia-lyase.
