摘要
目的探索复方壳多糖组织工程皮肤去牛血清清蛋白化的洗涤方法及对其单向混合淋巴细胞反应的影响。方法从头制备复方壳多糖组织工程皮肤,ELISA法检测不同洗涤条件下外渗液中牛血清清蛋白的浓度,然后检测在最优洗涤条件下组织工程皮肤单向混合淋巴细胞反应的强度。结果当洗涤间隔时间为3h,洗涤次数为2次时能使外渗液中牛血清清蛋白的浓度降到50ng/mL以下;按该方法洗涤后,组织工程皮肤的单向混合淋巴细胞反应强度显著降低。结论复方壳多糖组织工程皮肤以3h×2次洗涤,能够显著降低其外渗液中牛血清清蛋白的浓度及其单向混合淋巴细胞反应的强度。
Objective To explore the method of removing bovine serum albumin from composite chitosan artificial skin,and to research the altering of one-way mixed lymphocyte reaction after removing. Methods After the bovine serum albumin was removed form composite chitosan artificial skin,bovine serum albumin in washings and one-way mixed lymphocyte reaction were detected. Results At'ter the composite chitosan artificial skin was washed twice for 3h,bovine serum albumin in washings could be degraded to under 50ng/mL,and its one-way mixed lymphocyte reaction could be attenuated significantly also. Conelusion Our method could degrade bovine serum albumin in washings to a rational level,and degrade composite chitosan artificial skin's one-way mixed lymphocyte reaction significantly.
