摘要
[目的]探讨羧甲基壳聚糖对乙酸型大鼠胃溃疡黏膜保护作用。[方法]40只SD大鼠随机分为4组,每组10只,分别为:正常对照组、胃溃疡模型组、硫糖铝组(0.6g·kg-1·bw-1)、羧甲基壳聚糖组(0.5g·kg-1·bw-1)。除正常组外,其他各组均建立乙酸损伤型大鼠胃溃疡模型,造模3d后,正常组及胃溃疡模型组用蒸馏水灌胃,硫糖铝组与羧甲基壳聚糖组分别按设计剂量灌胃。治疗14d后处死大鼠,计算溃疡面积,测定胃黏膜SOD活性和MDA含量、并进行HE和免疫组织化学染色,观察VEGF,PCNA表达情况。[结果]与模型组相比,羧甲基壳聚糖组及硫糖铝组溃疡面积缩小,SOD活性高增加、MDA含量降低,VEGF,PCNA表达增加,差异有统计学意义(P﹤0.05)。[结论]羧甲基壳聚糖能增强乙酸型胃溃疡黏膜抗氧化能力,促进溃疡部位细胞增殖及VEGF,PCNA表达,加速胃溃疡愈合。
[Objective] To explore the protection of Carboxymethyl-chitosan (CM-chitosan) aginest gastric mucosa of gastric ulcer induced by acetic acid in rats. [Methods] 40 SD (Sprague-Dawley) rats were randomly divided into four groups: normal group, model group, sucralfate group and CM-chitosan group. Except normal group, the mede of gastric ulcer injured by acetic acid rats in other three groups was established. After establishing for 3 days, rats in normal group and model group were lavaged with distilled water, and in sucralfate group and CM-chitosan group were lavaged according to related dosage predesigned. The rats were sacrificed after treatment for 14 days. The ulcer size was measured. SOD in mucosa and MDA contents were detected, and conducted with immunohistoehemical stain and HE to observe the expression status of VEGF and PCNA. [Results] When compared with the model group, the ulcer size of rats in sucralfate group and CM-chitosan group showed to be deflated, and the activity of SOD significantly increased, the content of MDA significantly decreased as well as the expression of VEGF and PCNA significantly increased (P 〈 0.05). [Conclusion] The CM-chitosan has antioxidant effect on gastric mucosa, which could improve the cell multiplication and expression of VEGF and PCNA to accelerate the healing of gastric ulcer.
