食管鳞状细胞癌组织中EphA2和EphrinA1的表达 被引量:3

摘要 目的:观察食管鳞状细胞癌(食管鳞癌)组织中EphA2和EphrinA1蛋白及mRNA的表达水平,分析它们与食管癌患者预后的关系。方法:应用组织芯片技术和免疫组织化学方法检测173例食管鳞癌组织和19例正常食管黏膜组织中EphA2和EphrinA1蛋白表达水平;用激光捕获显微切割(LCM)技术和RT-PCR检测食管鳞癌组织中EphA2和EphrinA1 mRNA的水平。结果:食管鳞癌组织中EphA2阳性表达率80.9%、EphrinA1阳性表达率为84.4%,均明显高于正常食管黏膜(P<0.05);EphA2和EphrinA1蛋白表达同食管鳞癌患者的年龄、肿瘤部位、肿瘤大小、组织学分级无关;EphA2蛋白的表达水平与食管鳞癌患者的性别、淋巴结转移有关(P<0.05),EphrinA1蛋白的表达与食管鳞癌患者的临床分期有关(P<0.05)。生存分析显示:EphA2和EphrinA1蛋白的高表达与食管鳞癌患者的生存期有关。结论:EphA2和EphrinA1蛋白的高表达提示食管鳞癌患者预后不佳。 Aim: To observe the protein and mRNA expression of EphA2 and EphrinA1 in esophageal squamous cell carcinoma (ESCCs) , and analyze their relation with prognosis of ESCC. Methods : Tissue mieroarray and immunohistochemistry techniques were used to detect the protein expression of EphA2 and EphrinA1 in both esophageal cancerous tissue and normal epithelial tissue; laser capture microdissection (LCM) and reverse transcription polymerase chain reaction ( RT- PCR) were used to study the mRNA expression of EphA2 and EphrinA1 in ESCC. Results: The protein and mRNA expres- sions of EphA2 and EphrinA1 in ESCC were markedly increased than those in the normal esophageal epithelia( P 〈 0.05 ) ; EphA2 and EphrinA1 protein expressions were not associated with age, tumor location,tumor size or histological grade; but EphA2 protein expression was significantly associated with the gender ( P 〈 0.05 ) and lymph node metastasis( P 〈 0.05 ) , and EphrinA1 protein expressions were significantly associated with clinieal stage( P 〈 0.05). Univariate analysis suggested that the protein overexpression of EphA2 and EphrinA1 was correlated with the survival time of patients with ESCC. Conclusion: The protein overexpression of EphA2 and EphrinA1 suggests poor prognosis for patients with ESCC.
