摘要
目的:观察戊四氮点燃癫大鼠海马中水通道蛋白4(AQP4)和水通道蛋白9(AQP9)表达的变化,探讨它们在癫发生发展中的作用。方法:SD大鼠35只,随机分为对照组5只、生理盐水组5只及实验组25只。实验组大鼠腹腔注射戊四氮,制备戊四氮点燃癫大鼠模型;生理盐水组腹腔注射相应剂量生理盐水;对照组未做处理。实验组大鼠分别于癫持续发作后3h、6h、12h、24h和3d,各取5只大鼠脑组织,免疫组织化学SP法进行AQP4、AQP9检测。结果:实验组大鼠AQP4和AQP9蛋白主要分布于海马CA1、CA3区的锥体细胞层、齿状回颗粒细胞层和多形细胞层;癫持续状态3h时,实验组大鼠海马AQP4和AQP9蛋白表达降低,6h时增大至正常水平,而后逐渐增强,至24h达高峰,再逐渐降低,至3d时恢复至正常水平。结论:AQP4和AQP9与癫的发生发展关系密切。
Aim :To investigate the changes of expression of aquaporin 4 (AQP4) and aquaporin 9 (AQP9) in hippocampus of epileptic rats kindled by pentylenetrazol. Methods:Thirty five healthy male SD rats were randomly divided into normal group, saline control group and epilepsy group. The rats of epilepsy group were subjected to establish epilepsy model by injection of pentylenetetrazol. At 3 h,6 h,12 h,24 h ,3d after SE,AQP4 and AQP9 in hippocampus were detected using immunohistochemistry. Results :AQP4 and AQP9 protein were expressed in CA1 ,CA3 regions and dentate gyrus of therats in epilepsy group. Compared with those of the other two control groups, the expression of AQP4 and AQP9 in epilepsy group were lower at 3 h after SE, andnormal at 6 h after SE,began to rise significantly from 12 h after SE, reached a peak at 24 b after SE, and recovered to normol level at the 3rd day. Conclusion : AQP4 and AQP9 play an important role in epilepsy.
