戊四氮点燃大鼠海马脑源性神经营养因子受体TrKB、P^(75NTR)表达的变化

摘要 目的:探讨戊四氮点燃大鼠海马脑源性神经营养因子(BDNF)受体TrKB、P75NTR表达的变化。方法:40只成年健康雄性SD大鼠随机分为实验组和对照组,实验组采用戊四氮点燃法制备癫模型,对照组不做处理。第2周及第4周,2组各取10只大鼠,采用免疫组织化学方法观察海马结构内TrKB、P75NTR的表达水平。结果:第2周及第4周,对照组大鼠海马CA1、CA3区锥体细胞层及齿状回和门区可见TrKB阳性神经元和纤维;CA1、CA3区锥体细胞层、齿状回及门区可见大量P75NTR阳性神经元。与对照组比较,实验组海马相应部位TrKB的表达升高(P<0.05),P75NTR的表达降低(P<0.05)。结论:在癫形成过程中,BDNF的2类受体TrKB、P75NTR可能参与了癫后海马异常苔藓纤维出芽及突触重建过程;2类受体可能相互拮抗。 Aim : TO explore the changes of TrKB, p^75NTR expression in hippocampus of rats kindled by pentyenetrazol. Methods:Forty healthy male rats were randomly divided into experimental group and control group. Rats in experimental group were kindled by pentyenetrazol. The expression of TrKB and p^75NTR in the hippocampus of the rats were determined by immunohistochemistry at Week 2 and Week 4. Results : Compared with those of the control at Week 2 and Week 4, p^75NTR expression in the inner molecular layer of the dentate gyrus, hillus and the mossy fiber layer of the area CA1, CA3 in the hippocampus decreased(P 〈 0.05 ), and TrKB expression in the hippocampus of the experimental rats increased( P 〈 0.05 ). Conclusion : TrKB and p^75NTR may be involved in the process of aberrant mossy fiber sprouting and synaptic plastic- ity in the hippocampus after seizures, and the roles of the two receptors may be antagonistic.
