摘要
目的:比较体外不同生长状态的晶状体上皮细胞(LECs)对药物敏感性的差异,为体外药物筛选设计提供参考。方法:用含体积分数为10%胎牛血清(FBS)的DMEM培养基调整LECs浓度,分为2组:未融合增生组(细胞浓度为4×104mL-1)和融合维持组(细胞浓度为4×105mL-1),分别放入24孔板中,每孔接种1mL。接种后12h,未融合增生组、融合维持组分别用含体积分数为20%、2%的FBS及含有不同处理因素(含有不同质量浓度5-氟尿嘧啶、丝裂霉素、肝素和地塞米松的DMEM培养基)的培养液培养。作用48h后用MTT比色法检测细胞增生情况。结果:5-氟尿嘧啶和丝裂霉素对未融合增生组和融合维持组的LECs均产生明显的浓度依赖性抑制作用,同一质量浓度药物作用下,未融合增生组的抑制率高于融合维持组(P<0.05)。肝素和地塞米松对未融合增生组LECs有较明显的浓度依赖性抑制作用(P<0.05),而对融合维持组的LECs几乎没有影响。结论:体外不同生长状态的LECs对不同药物及剂量敏感性不同。5-氟尿嘧啶和丝裂霉素抑制LECs的增生作用强,但对处于融合维持状态的LECs毒性也明显;肝素和地塞米松对LECs有一定的增生抑制作用,对处于融合维持状态的LECs毒性低。
Aim : To compare the sensibility of bovine lens epithevial cells(BLECs) to the different drugs in different growth state in vitro so as to provide data for the medicine selection. Methods:The concentration of cells was adjusted with DMEM medium supplemented with 10% FBS and the cells was cultured in 24-well plates and divided into two groups. The first group was added 4 × 10^4cells in 1 mL per well and the second group was 4 × 10^5 cells. After 12 h, the medium of the first group was changed : the medium was supplemented with 20% FBS and the different concentration of the drugs, and the second, with 2% FBS and the different concentration of the drugs. Each experimental group had a control group. After 48 h, the inhibition rate was tested by MTT method. The drugs were 5-FU, MMC,heparin and dexamethasone. Results: 5-FU and MMC inhibited the proliferation of BLECs depending on the concentration in the first group and the second group. The inhibition rate in the first group was higher than that in the second group. Heparin and dexamethasone inhibited the prolifer- ation of BLECs obviously in the first group ,but almost had no effects on the second group. Conclusion: There are different sensibility to the different drugs for BLECs in different growth state in vitro. 5-FU and MMC can strongly inhibit the prolif- eration of BLECs in different growth state.
