垂体腺瘤组织中ASPP2 mRNA和Ki-67蛋白的表达

摘要 目的:研究垂体腺瘤组织中ASPP2基因mRNA和Ki-67抗原的表达情况。方法:收集垂体腺瘤手术标本71例(其中侵袭性垂体腺瘤36例,非侵袭性垂体腺瘤35例;病理分型包括生长激素腺瘤14例,泌乳素腺瘤13例,促肾上腺皮质激素腺瘤5例,促性腺腺瘤7例,无功能腺瘤10例,多激素腺瘤22例),采用RT-PCR方法检测垂体腺瘤标本中ASPP2基因mRNA的表达情况,采用免疫组织化学方法检测垂体腺瘤石蜡切片中Ki-67抗原的表达情况,并分析二者的相关性。结果:71例垂体腺瘤手术标本均有ASPP2基因mRNA和Ki-67表达,不同病理类型间ASPP2基因mRNA表达差异无统计学意义(P>0.05);但不同病理类型间Ki-67 LI差异有统计学意义(P<0.05)。非侵袭垂体腺瘤和侵袭性垂体腺瘤组织中ASPP2 mRNA的表达和Ki-67 LI差异均有统计学意义(P<0.05);AS-PP2基因表达水平与Ki-67 LI呈负相关(r=-0.256,P<0.05)。结论:人垂体腺瘤组织中ASPP2基因的表达与其细胞增殖活性和侵袭性行为有关,可以作为判断垂体腺瘤侵袭性和预测复发的有效临床检测指标。 Aim : To study the expression of ASPP2 mRNA and Ki-67 antigen in pituitary adenomas. Methods : A total of 71 cases of pituitary adenomas were collected( including 36 cases of invasive pituitary adenoma and 35 cases of noninvasive pituitary adenomas. Patho-category:13 cases of PRL adenoma,14 cases of GH adenoma,5 cases of ACTH adenoma,7 cases of gonadotroph adenoma,10 cases of nonfunctional adenoma, and 22 cases of multi-hormone adenoma). RT-PCR was used to detect the expression of ASPP2 mRNA in samples. Immunochemistry technique was applied to examine Ki-67 ex- pression in the paraffin section of samples. Results : ASPP2 mRNA and Ki-67 protein were detectable in all samples of pituitary adenomas. There was no statistical difference in ASPP2 mRNA among different pathological type (P 〉 0.05 ) ,however there was statistical difference in Ki-67 protein among different pathological type( P 〈 0.05 ). The ASPP2 mRNA and Ki- 67 protein between invasive pituitary adenoma and noninvasive pituitary adenomas had significant differences( P 〈 0. 05). Correlation between ASPP2 and Ki-67 LI was negative ( r = - 0. 256, P 〈 0.05 ). Conclusion : The expression of ASPP2 mRNA in human pituitary adenoma tissue is related with the proliferation and invasion of tumor. They might be a valuable clinical marker in prediction of the invasion of pituitary adenomas.
