林肯链霉菌原生质体制备条件的研究

Study on Protoplast Preparation of Streptmyces Lincolsis

对林肯链霉菌SL98原生质体制备条件进行了探索。其原生质形成最佳培养基中含甘氨酸浓度为0.4%,蔗糖浓度为34%;酶解条件是采用溶菌酶浓度4mg/mL,菌龄72h,酶解时间5h,酶解温度为30℃。其最高的原生质体的制备率达16.3%。

The conditions of producing and regenerating protoplast of Streptmyces Lincolsis were discussed. Concentrations of glycine and sugar in optimal substrate were 0.4% and 34% respectively. The optimum conditions for protoplast production of streptomyces Lincolsis were as follows： culturing 72 h with lysine enzyme 4mg/mL, lysine time 5 h at 30℃. The producing rate of protoplast can reach 16.3% under such conditions.
