摘要
目的研究人参水提物(WEG)对四氢吡啶离子(MPP+)诱导SH-SY5Y细胞凋亡的影响。方法体外培养SH-SY5Y细胞,建立MPP+致SH-SY5Y细胞凋亡的模型,同时给予WEG干预。应用MTT比色法检测细胞活力;Hoechst33258染色观察细胞形态学变化,流式细胞仪检测细胞凋亡率。结果0.5mmol·L-1MPP+作用于SH-SY5Y细胞60h,MTT值降低到(51.97%±2.46%)(P<0.01),Hoechst 33258染色可见明显的颗粒状和固缩状荧光,凋亡率增加到(40.52%±1.68%)(P<0.01),而WEG则能明显改善SH-SY5Y细胞的凋亡特性,显著升高其MTT值,降低凋亡率(P<0.05)。结论人参水提物对MPP+诱导的SH-SY5Y细胞凋亡具有显著的保护作用。
Objective To investigate the protective effect of water extracts of Ginseng on SH-SY5Y cell apoptosis induced by MPP^+ in culture.Methods MPP+ induced SH-SY5Y cell apoptosis model was established.Water extracts of Ginseng was added into the culture to test its protective effect.The cell viability was measured by MTT.To test the morphology of SH-SY5Y,cells were observed by Hoechst 33258 staining and apoptotic rate was analyzed by FCM.Results The cell activity decreased remarkably induced by 0.5 mmol·L^-1 MPP^+ for 60 h(P〈0.01)and the cells were positively stained with fluorochrome Hoechst 33258,presenting apoptotic rate to(40.52%±1.68%),(P〈0.01),However,water extracts of Ginseng obviously improved MTT value,and reduced cell apoptotic rate.Conclusion Water extracts of Ginseng have marked protective effect on SH-SY5Y cell apoptosis induced by MPP^+.
