乳腺癌组织中血管内皮生长因子与转录因子共活化物PGC-1α的表达

Expression of VEGF and PGC-1α in breast carcinoma

摘要 目的探讨乳腺癌组织中血管内皮生长因子(VEGF)和转录因子共活化物过氧化物酶体增殖物激活受体γ辅激活因子-1α(PGC-1α)的表达情况及其与癌组织生长、浸润及转移的关系。方法应用免疫组织化学和RT—PCR技术检测26例乳腺浸润性导管癌和9例正常乳腺组织中VEGF、线粒体DNA(mtDNA)水平及转录因子共活化物PGC-1α的表达。结果乳腺癌组织中VEGF蛋白及mRNA表达水平明显高于正常乳腺组织,而线粒体及转录因子共活化物PGC-1α的mRNA的表达水平则低于正常乳腺组织。结论乳腺癌组织中VEGF蛋白及mRNA表达显著上调,而PGC-1αmRNA表达显著下调。乳腺浸润性导管癌组织生长、浸润及转移可能与VEGF表达上调及PGC-1α表达降低有关。 Objective To explore the expression of VEGF and PGC-1α in breast carcinoma and its relationship to the tumor proliferation,infiltration and rmetastasis. Methods Immunohistochemistry and RT- PCR were used to examine the expression of VEGF, mtDNA and PGC-1α in 26 infitrating ductal breast carcinoma examples and 9 normal breast tissues respectively. Results The expression of VEGF protein and mRNA in the breast carcinoma was obviously higher than that in the normal tissues, but the mRNA expression of mtDNA and PGC-1α was higher in normal tissues. Conclusion The expression of VEGF up-regulates and PGC-1α down-regulates in the breast cancer tissue. Lower level of PGC-1α may play a role in the breast tumor cells proliferation, infiltration and metastasis.
