大鼠颈总动脉重度狭窄模型建立与超声对其的评价 被引量:5

摘要 目的建立大鼠颈总动脉(CCA)重度狭窄模型,并探讨采用高频彩色多普勒血流显像(CDFI)评价其狭窄程度。方法将19只SD大鼠随机分为两组,假手术组11只,CCA狭窄组8只。采用针控线栓法制作大鼠CCA狭窄模型。术后3 h以高频CDFI检测CCA内径、收缩期峰值流速(PSV)、舒张期峰值流速(EDV)、PSVCCA/颈内动脉PSV(PSVICA)及阻力指数(RI),评价CCA狭窄程度。术后6 h,在显微镜直视下观察CCA血管内壁情况。结果假手术组原始管径为(0.65±0.07)mm,PSVCCA中位数为101.1 cm/s,EDVCCA中位数为13.4 cm/s;PSVCCA/PSVICA为1.36;狭窄组大鼠残余管径为(0.18±0.06)mm,PSVCCA中位数为334.2 cm/s,EDVCCA中位数为98.4 cm/s,PSVCCA/PSVICA为6.43。两组相应指标比较,差异有统计学意义(P<0.05)。两组大鼠超声检查结果与显微镜直视下的观察结果完全符合。结论采用高频CDFI可准确评价线栓法制作的大鼠CCA重度狭窄模型的狭窄程度。 Objective To develop a rat model of severe common carotid artery (CCA) stenosis and to evaluate the stenotic degree with high-frequency color Doppler uhrasonography. Methods Nineteen SD rats were randomly allocated into 2 groups: a sham-operation group ( n = 11 ), and a CCA stenosis group (n =8). A CCA stenosis model was induced by suture method. The CCA internal diameter, peak systolic velocity ( PSV ) , end diastolic velocity ( EDV ) , PSVCCA/PSV in internal carotid artery ( PSVICA ) , and resistance index (RI) were measured by the high-frequency color Doppler ultrasonography at 3 hours after the procedure, and the stenotic degree of CCA was evaluated. The CCA walls were observed directly under the microscope at 6 hours after the procedure. Results The original diameter in the sham-operation group was 0. 65±0. 07 mm. The median PSVCCS was 101.1 cm/s, the median EDVCCS was 13. 4 cm/s, and the PSVCCA/PSVICA Was 1. 36. The residual lumen was 0. 18±0. 06 mm in the CCA stenosis group, and the stenosis rate was 70±10%. The median PSVCCA was 334. 2 cm/s, the median EDVCCA was 98.4 cm/s, and PSVCCA/PSVICA was 6. 43. There were statistically significant differences between the corresponding indexes in both groups (P 〈 0. 05 ). The results of ultrasonography in both groups were completely in accordance with those observed directly under the microscope. Conclusion The stenotic degree of the rat model of severe CCA stenosis induced by a suture method can be accurately evaluated by using high-frequency color Doppler ultrasonography.
