摘要
目的建立一种新型化学发光测定一氧化氮方法。方法用氩气将全血样品中一氧化氮(NO)排出进入发光仪中鲁米诺-过氧化氢(H2O2)体系,NO与H2O2反应氧化鲁米诺产生发光。用碘化钾(KI)在酸性环境中使部份NO代谢产物还原为NO,增加方法的灵敏度。用EDTA为络合剂,无水乙醇为去泡剂,去除金属离子和血气泡的干扰。结果NO含量与发光强度呈正相关,120~1000pmolNO含量范围内有良好的线性关系(r=0.998)。全血一氧化氮浓度为6.55μmol/L时,批内变异系数(CV)为8.71%。结论该方法用于全血NO测定,灵敏、特异,可避免液体介质中各种因素干扰。
Objective To establish a method of chemiluminescence for measuring blood nitric oxide (NO). Methods Nitric oxide in blood was driven out and introduced to chemiluminescence detector with a stream of argon. NO reacted with H 2O 2 to form peroxynitrite (ONOO -). Which was a stronger oxidant and could oxidize luminol to generate chemiluminescence. Results There was a linear relationship between the amount of NO and the chemiluminescence intensity over the range of 120 pmol~1 000 pmol of NO. Conclusions The assay was described in this paper was rapid, sensitive and specific and the interfere factors in liquid could be avoided.
