摘要
目的建立一种测定乳酸的新方法。方法采用乳酸氧化酶氧化乳酸产生丙酮酸和H2O2,然后应用Trinder显色反应测定血浆中乳酸。结果该方法乳酸氧化酶用量为200U/L,酶反应时间10分钟,线性范围可达11mmol/L,变异系数(CV)为2.31%~3.25%,回收率为98.4%~101.0%,与法国生物-梅里埃试剂盒比较具有良好的相关,r=0.985,Y=0.987X+0.040,40例健康人血浆乳酸值为0.60~2.52mmol/L(x±2s)。结论乳酸测定用乳酸氧化酶法测定乳酸具有快速、简便、灵敏、可靠等优点,适合临床应用。
Objective To develop a method of lactate oxdase for measuring lactate in plasma. Methods Lactate was oxidized by lactate oxidase to pyruvate and hyarogen peroxide, which in presence of peroxidase reactsed with trinder forming a compound. The colour in tensity of which was propotional to the concertration of lactate in the examined sample. Results The assay contained 200 U/L of lastate oxidase, and enzyme reaction time was 10min. The linearlity of tast was up to 11mmol/L. The withinrun coefficients of variation for controls at the concentrations were 2.31% to 3.25%. The recoveries ranged from 98.4% to 101.0%. Comparing this method with biomerieux reagents method, we obtained the regression equation, r =0.985, Y =0.987 X +0.024. The reference inteval ( ±2 s ) was 0.60 to 2.52 mmol/L n =40. Conclusions This method is simple, sensitive, and suitable for clinical application.
